Priming of the system is used to expel air from various tubings etc. See HPLC manual, for example:

Priming and Purging the System
When the solvents have been exchanged or the pumping system has been
turned off for a certain time (for example, overnight) oxygen will re-diffuse
into the solvent channel between the solvent reservoir, vacuum degasser
(when available in the system) and the pump. Solvents containing volatile
ingredients will slightly lose these. Therefore priming of the pumping system
is required before starting an application.

The pump should never be used for priming empty tubings (never let the pump run dry). Use a syringe to draw enough solvent for completely filling the tubings to the pump inlet before continuing to prime with the pump.

The principle is exactly the same: you need to take somewhat more blood and to prime the tubings before analysis. If the analyzed blood sample is, for example, 1 ml, and the volume of the tubings to be primed is also 1 ml, you need 2 ml of blood total

Notes to answerer
Asker: Natalie or Boris, would you mind elaborating? The only priming/primer that I am familiar with in a lab environment are those related to PCR, ELISA, and bacterial culture. I could not find any "system priming" for bioavailability/bioequivalence research. Thank you for your time.

Asker: P.S. The methods used here are: high performance liquid chromatography combined with mass spectrometry (HPLC-MS/MS).

Asker: Thank you Natalie. I am very familiar with this concept of priming. I prime my gas-powered tools before starting them. But how are bio-samples of whole human blood are used for priming a chromatography equipment?
