Manche sagen, das Elend habe 1987 begonnen, irgendwo in Manhattan. Dort zimmerten einige oberschlaue Investmentbanker der Firma Drexel Burnham Lambert das erste Kreditderivat zusammen - die Mutter all jener collateralized debt obligations, die zurzeit die Bilanzen großer Unternehmen im Wochentakt implodieren lassen.

Nun weiß man: Die Katastrophe begann bereits vier Jahre früher, in einer kleinen Mailänder Espressobar. Dort, im Cafe "L'inizio della fine" machte ein Mann jene folgenschwere Entdeckung, die ein Vierteljahrhundert später das gesamte westliche Wirtschaftssystem zum Einsturz bringen sollte.
Der Mann hieß Howard Schultz und kostete in Mailand seinen ersten gescheiten Espresso. Der Amerikaner kapierte plötzlich, dass Kaffee nicht wie abgestandenes Brackwasser schmecken muss. Nach seiner Rückkehr in die USA erfand Schultz Starbucks, heute der größte Latte-Dealer des Planeten.
Lange schienen Schultz' Shops ein Segen für die gesamte Menschheit zu sein. Erstmals gab es überall lauwarmen Kaffee zu überzogenen Preisen. Besonders gestresste Investmentbanker und Wertpapierhändler wussten die belebende Wirkung des kräftigen Gebräus aus Seattle zu schätzen.
Und so nahm das Verhängnis seinen Lauf.
Eine Studie der Universität Durham hat jetzt aufgedeckt, dass übermäßiger Kaffeekonsum zu Halluzinationen führen kann. Ab der siebten Tasse (330 Milligramm Koffein) fangen Bohnenjunkies an, Dinge zu sehen, die überhaupt nicht vorhanden sind.
Flüssiges Rohypnol im Pappbecher
Sieben Tassen, das klingt nach viel. Ist es aber nicht. Die Durhamer Probanden bekamen nämlich dünnen Filterkaffee verabreicht. Ein Triple-Venti-Cappucino, wie ihn Investmentbanker bevorzugen, hat natürlich deutlich mehr Wumms. Oder anders gesagt: Wenn ein Tässchen Jacobs Krönung einer Aspirin entspricht, dann wirkt ein Eimer von Starbucks wie eine Kurpackung Rohypnol.
Die Merrills und Lehmans kübelten sich täglich vier bis fünf Ventis hinter die Binde, das Äquivalent von 40 bis 50 Tassen normalen Kaffees. Mit dieser Menge Koffein könnte man einen Blauwal vom Wegdösen abhalten. Folglich halluzinierten die Banker drauf los, als hätten sie ein ganzes Löschblatt Sunshine-Acid durchgekaut. Sie erblickten Vermögenswerte, die sonst niemand sah - psychedelische CDOs, trippige Subprime-Hypotheken, abgefahrene Plain Vanilla Swaps.
Wenn man schon nach sieben Tassen blaue Elefanten phantasiert, sieht man nach 50 Bechern natürlich noch viel wahnwitzigeres Zeug: Zum Beispiel Immobilien in Kansas, die ständig im Wert steigen. Oder Anleihen auf abgeklungene Plutonium-Brennstäbe, die ein AAA-Rating aufweisen.
Mit der Zeit wurde der Kaffeekoller zur Gruppenpsychose. Die Banker waren derart mit Tres Rios Costa Rica Bella Vista zugeballert, dass sie begannen, sich ihre Halluzinationen gegenseitig zu verkaufen. Kurz darauf flog uns alles um die Ohren.
Warum stürzt der Kaffeepreis nicht ab?
Das ganze klingt prima facie verrückt, doch das Arabica-Theorem ist über jeden Zweifel erhaben. Das beweist ein Blick auf jene Länder, die von der Finanzkrise am schlimmsten verheert wurden. Die Starbucks-Nation USA? Zerschmettert! Die ständig "Koepi" süffelnden Südkoreaner? Beim Abdecker! Und womit hielten sich wohl die Isländer während der endlosen Winter warm? Genau, mit viel heißem Macchiato, den sie mit ihren Kaupthing-Kreditkarten bezahlten.
Mutmaßlich kommt demnächst alles noch schlimmer. Denn während sämtliche Rohstoffpreise in den vergangenen Monaten in den Orkus getrudelt sind, blieb der Preis für Kaffeebohnen an den Terminbörsen stabil. Irgendjemand da draußen säuft also nach wie vor Cappuccino aus viel zu großen Bechern.
Sobald uns der Turbo-Kaffee komplett ruiniert hat, kommen dann die Chinesen und kaufen die Überreste unseres Turbo-Kapitalismus auf. Das plant die Führung der Volksrepublik schon seit langem. Oder warum sonst, fragt man sich, trinken die Chinesen ihren Tee aus ganz kleinen Tassen?

Capitalism is over, guilty parties are sought: Were they investment bankers or Americans addicted to credit? Scientists now suggest a different culprit: the Large Triple-Shot Columbia Nariño Supremo Cappucino.
Some say that the misery began in 1987, somewhere in Manhattan. There a few really clever investment bankers at Drexel Burnham Lambert carpentered the first credit derivative – the mother of all those collateralized debt obligations that, at the moment, are causing a weekly drumbeat of imploding balance sheets at large businesses.
Coffee beans: After the seventh cup, according to researchers at Durham University, hallucinations can arise. This problem affects the finance- and creative industries more than any others.
Now we know: The catastrophe began four years earlier, in a small espresso bar in Milan. There, in the Café “L'inizio della fine,"a man made a fateful discovery that a quarter century later would bring the entire western economic system to its knees.
The man was named Howard Schultz, and in Milan did he taste his first decent espresso. The American suddenly understood that coffee didn’t have to taste like stale, brackish water. After his return to the States, he established Schultz Starbucks, today the largest latte-pusher on the planet.
For a long time, Schultz;s shops appeared to be a boon for the whole of humankind. For the first time ever, lukewarm coffee at exorbitant prices was available everywhere. Over-stressed investment bankers and securities dealers came to value the stimulating effect of this powerful concoction from Seattle.
And thus did the disaster chart its course.
A study at Durham University has now revealed that excessive consumption of coffee can lead to hallucinations. After the seventh cup (330 milligrams of caffeine), bean junkies begin seeing things that are absolutely not there.

Seven cups. That sounds like a lot. But it isn’t. In other words, the Durham human guinea pigs were served thin, filter-coffee. Of course, a triple venti cappuccino, as investment bankers prefer, has significantly more kick. In other words: If a little cup of Jacobs Kronung coffee corresponds to an aspirin, then a bucket of Starbucks is like a pack of the tranquilizer Rohypnol.
The Merrills and Lehmans knocked back four or five ventis every day, the equivalent of 40 to 50 cups of normal coffee. With this amount of caffeine, someone could keep a blue whale from dozing off. As a result, the bankers started to hallucinate as if they had chewed through an entire blotting-paper of LSD. They perceived assets that no one else saw – psychedelic CDOs, tripping subprime mortgages, far-out plain-vanilla swaps.
If you fantasize blue elephants after seven cups, then after 50 you’re going to see things that are even more insane. For example, real estate in Kansas that continually rises in value. Or bonds based on depleted plutonium fuel-rods that showed a AAA rating.
Over time, the coffee rage became a group psychosis. The bankers were so hammered with Tres Rios Costa Rica Bella Vista that they began selling their hallucinations to each other. Shortly thereafter, everything blew up in our faces.
Why didn’t the price of coffee tank?
The whole thing sounds crazy on the face of it, yet the Arabica Theory is beyond any doubt. A glance at those countries that were most seriously devastated by the finance crisis proves as much. The Starbucks nation USA? Shattered! The constantly Koepi-tippling South Koreans? At the knackers. And how are the Icelanders going to keep warm during their interminable winter? Exactly, with a very hot Macchiato, which they pay for with their Kaupthing-bank credit cards.
Presumably, things will get even worse. Because during recent months, as the collective prices of commodities went all to hell, the price of coffee beans on the futures exchanges remained firm. So someone out there, now as before, is chugging Cappuccino from mugs that are way too big.
As soon as the turbo-coffee has completely ruined us, along come the Chinese and buy up the rest of our turbo-capitalism. That’s what the leadership of the People’s Republic had planned all along. Otherwise, one must ask, why do the Chinese drink their tea from such small cups?

Durchbruch in der Biomedizin: US-Forschern ist es an Mäusen gelungen, Körper- in Stammzellen zu verwandeln - ohne dabei Gentechnik einzusetzen. Sie schafften das Kunststück, indem sie die Zellen mit einem Protein-Gemisch behandelten.
Ein Arztbesuch im Jahr 2020 könnte so aussehen: Mit einem kleinen Schaber kratzt der Doktor vom Arm des Patienten ein wenig Haut ab und bittet ihn, nach einer Woche wiederzukommen. Dann werde man seinen Diabetes heilen. In der Zwischenzeit kommt seine Zellprobe in eine Kulturschale, auf die die Arzthilfe ein Gemisch aus chemischen Substanzen schüttet. Nach wenigen Tagen haben sich die Zellen in Stammzellen verwandelt, die Arzthilfe schüttet einen weiteren Chemie-Cocktail darauf - und am Ende sind in der Kulturschale fertige Bauchspeicheldrüsenzellen.
Noch ist es nicht soweit, aber an der Umwandlung von Körper- zu Alleskönner-Stammzellen wird derzeit intensiv gearbeitet. In den vergangenen Monaten verbuchten mehrere Arbeitsgruppen wichtige Etappenerfolge bei der Verbesserung der Methode: Die Reprogrammierung gelang mit immer weniger eingeschleusten Genen, ohne Viren und letztlich konnten die Gene sogar nach getaner Arbeit wieder aus den Zellen entfernt werden.
Alle diese Methoden erforderten jedoch eine genetische Veränderung der Zellen - was Risiken birgt und einer zukünftigen medizinischen Anwendung im Wege steht. Der Königsweg wäre daher die Reprogrammierung der Zellen mit so wenig Eingriffen wie möglich - ohne Gene und nur mit Hilfe "kleiner Moleküle", wie es der deutsche Stammzellforscher Hans Schöler vom Max-Planck-Institut für molekulare Biomedizin formuliert. Mit anderen Worten: ein chemisches Gemisch, das man nur noch auf die Zellen kippt und sie sich daraufhin in Stammzellen verwandeln.
Reprogrammierung ohne Gentechnik
Das ist nun US-amerikanischen Forschern um Sheng Ding vom Scripps Research Institute im kalifornischen La Jolla erstmals an Mäusezellen gelungen - oder zumindest fast: Wie die Forscher im Fachmagazin "Cell Stem Cell" berichten, benutzten sie zwar keine "kleinen", sondern große Proteinmoleküle - aber das bahnbrechende an ihrer Arbeit ist, dass sie erstmals das Erbgut der Zellen selbst nicht mehr verändern mussten.
Die Reprogrammierung von Zellen gelang vor drei Jahren erstmals dem japanischen Zellforscher Shinya Yamanaka - zuerst an Mauszellen, später auch beim Menschen. Er hatte gezeigt, dass durch die Einschleusung bestimmter Gene spezialisierte Zellen in ihren embryonalen Ursprungszustand zurückversetzt werden können. Diese sogenannten induzierten pluripotenten Stammzellen (iPS) stehen in ihren Fähigkeiten embryonalen Stammzellen (ES) in nichts nach. Die neuen zellulären Wunderkinder waren geboren - und zudem waren sie noch ethisch unproblematisch, weil zu ihrer Herstellung keine Embryonen benötigt werden.
Bislang hatten alle Arbeitsgruppen Reprogrammier-Gene, üblicherweise die Gene Oct4, Klf4, Sox2, c-Myc, direkt in die Zellen eingeschleust. Ding und seine Kollegen hingegen nahmen die Proteine, in die die Gene normalerweise in den Zellen übersetzt werden. Weil diese Proteine große Moleküle sind, mussten die Wissenschaftler mit einem Trick nachhelfen, damit sie von den Zellen aufgenommen wurden: Sie hängten mehrere Moleküle der Aminosäure Arginin an die Proteine. Sozusagen als Eintrittskarte: Der Arginin-Schwanz befördert die Aufnahme großer Moleküle in die Zelle. Die Proteine selbst ließen die Forscher von Bakterien produzieren.
Ding und sein Team verwendeten für ihre Versuche Hautzellen aus Maus-Embryonen. Diese behandelten sie viermal mit dem Cocktail aus Oct4-, Klf4-, Sox2- und c-Myc-Proteinen. Hans Schöler, dessen Team die Hautzellen geliefert und die Stammzellen begutachtet hatte, sagte im Gespräch mit SPIEGEL ONLINE, die viermalige Behandlung sei nötig gewesen, weil die Zellen sonst wieder in ihren differenzierten Zustand zurückfallen würden.
"Wir müssen uns nicht mehr mit Mutationen beschäftigen"
Nach etwa 35 Tagen war es dann soweit: die Mauszellen waren reprogrammiert. PiPS nennen die Forscher ihre neuen Kreationen: Protein induzierte pluripotente Stammzellen. Den gängigen Test auf Pluripotenz erfüllten sie. In Mäuseembryonen injiziert, trugen sie zur Bildung der drei Keimblätter bei, aus denen sich im Laufe der Embryonalentwicklung alle Organe und Gewebe bilden.
Schneller, sicherer und einfacher sei ihre neue Reprogrammier-Methode schreiben Ding und seine Kollegen in "Cell Stem Cell". Selbst wenn noch große Proteinmoleküle zur Reprogrammierung nötig sind, das Hauptproblem der Zell-Reprogrammierung scheint nun gelöst, wie Hans Schöler zuversichtlich sagt: "Wichtig ist, dass wir nun eine biochemische Reprogrammierung ohne DNA in den Händen haben. Das heißt, dass wir uns nicht mit potentiellen DNA-Insertionen und daraus resultierenden Mutationen beschäftigen müssen."
Der Arztbesuch der Zukunft ist damit wieder etwas näher gerückt. Nun müssen die Forscher am anderen Ende der Kette weitere Fortschritte leisten: Für jeden gewünschten Zelltyp muss die jeweilige Mixtur gefunden werden, die man auf die hergestellten Stammzellen schütten muss, damit sie sich darin verwandeln. Beispielsweise gelang es Forschern schon, aus iPS-Zellen Nerven- und Herzmuskelzellen zu züchten.
Eine Frage allerdings bleibt offen: Werden die Stammzellforscher, die liebevoll von "Ipsen" sprachen, wenn sie iPS-Zellen meinten, die zellulären Neulinge nun konsequenterweise auch "Pipse" nennen?

Breakthrough in biomedicine: US researchers have succeeded in transforming body cells into stem cells—without employing genetic engineering. They accomplished this feat by treating the cells with a blend of proteins.
In the year 2020, a visit to the doctor could go like this: The doctor scratches a bit of skin from the patient’s arm with a small scraper, and asks him to return in a week. At that time, he’ll have his diabetes cured. Meanwhile, his cell sample goes into a Petri into which dish the doctor’s assistant pours a mix of chemical substances. After a few days, the cells have transformed themselves into stem cells, and the assistant pours another chemical cocktail onto them—and ultimately there are pancreas cells in the Petri dish, all ready to go.
Things have not yet progressed so far, but the transformation of body cells into general-purpose stem cells is being worked on intensively. In past months, several teams have recorded important interim successes in improving the process: Reprogramming has succeeded with ever fewer genes inserted, without [resorting to] viruses, and ultimately the genes could even be extracted again from the cells after the work was finished.
However, all these procedures demanded a genetic alteration of the cells—something that concealed risks and stands in the way of a future medical application. Therefore, the ideal way would to reprogram cells with as little intrusion as possible—without genes and with only the help of “small molecules,” as the German stem-cell researcher Hans Schöler of the Max-Planck Institute for Molecular Biomedicine put it. In other words: a chemical cocktail, that one merely pours onto the cells with the result that they turn themselves into stem cells.
Reprogramming without genetic engineering
That’s now what US researchers um Sheng Ding of the Scripps Research Institute in La Jolla, California have in mouse cells achieved for the first time—or almost, at least: As the researchers in the professional journal Cell Stem Cell” recount, instead of “small” protein molecules, they actually used large ones. But the pioneering aspect of their work is that, for the first time, they didn’t have to alter the genetic makeup of the cells.
Three years ago the Japanese cell researcher Shinya Yamanaka first succeed in the reprogramming of cells—in the beginning on mouse cells, later also on human beings. He had shown that through the insertion of certain genes, specialized cells could be restored to their original embryonic state. These so-called induced pluripotent stem cells are the equal of embryonic stem cells (ES) in their capabilities. The new cellular prodigies were born—and furthermore were ethically unproblematic because no embryos were required for their production.
Until now, all [stem-cell research] teams had inserted reprogramming genes—usually the genes Oct$, Klf4, Sox2, c-Myc—directly into the cells. In contrast, Ding and his colleagues used the proteins normally expressed by the genes in the cells. Because these proteins are large molecules, the scientists had to employ a trick so that they would be accepted by the cells: They hung several molecules of the amino acid arginine on the proteins. The entry pass, in a manner of speaking: The arginine tail promotes the admittance of larger molecules to the cells. The researchers enlisted bacteria to produce the proteins themselves.
Ding and his team used skin cells from mouse embryos for their experiments. They treated the cells four times with the cocktail consisting of Oct4-, Klf4-, Sox2- and c-Myc proteins. Hans Schöler, whose team had supplied the skin cells and had examined the stem cells, said in a conversation with SPIEGEL ONLINE, the four treatments were necessary because otherwise the cells would have returned to their differentiated state.
“We must no longer tinker with mutations”
After about 35 days, the time had come: the mouse cells were reprogrammed. The researchers called their new creations PiPS—protein induced pluripotent stem cells. The researchers performed the common test for pluripotency. Injected into mouse embryos, they contributed to the formation of the three germ layers from which all organs and tissues grow during development of the embryo.
As Ding and his colleagures write in “Cell Stem Cell,” their new reprogramming method is faster, safer, and simpler. Even though large protein molecules are still necessary for reprogramming, the main problem of cell reprogramming now appears to be solved, as Hans Schöler said confidently: It’s important that we now have in hand biochemical reprogramming without DNA. That is, that we don’t have to concern ourselves with potential DNA insertions and the resulting mutations.”
With this technology, the doctor’s visit of the future has moved somewhat closer. Now researchers must make additional progress at the other end of the chain: For every cell type of interest, the respective mixture must be found to pour over manufactured stem cells so that they transform themselves into the kind of cell sought. For instance, researchers have already succeeded in cultivating nerve and heart-muscle cells from iPS cells.
However, one question remains open: will the stem-cell researchers, who fondly speak of “ipses,” when they mean iPS cells, now also consistently call the cellular novices “pipses?”
